Capsaicin (in chili peppers), ptperine I in black pepper), and anandamide act on the capsaicin receptor (vanilloid receptor 1. VR1) on tow-threshold heat-sensing neurons with small to medium diameter, which arc present throughout the oral cavity. VR1 is a cation channel, which would otherwise be activated by exposure to temperatures over 43°C. Capsaicin does not traverse the channel itself, but binds to an intracellular moiety and opens the channel for cations. The ensuing influx of sodium and calcium ions then leads to the more or less persistent activation of the nociceptor neuron (Tominaga and Julius. 2000), Acids can activate the receptor directly and also increase its responsiveness to capsaicin and heat (Jordt et at., 2000). It is of note that capsaicin docs not directly damage cells, the considerable potential for perceived discomfort notw ithstanding. It is only the response to neural stimulation that induces potentially harmful responses. Because they lack a capsaicin-responsive VR1. birds can tolerate large doses of capsaicin (e.g. from seeds laced with it), while mammals (e.g. squirrels) cannot.

While capsaicin has no significant effect on gastrointestinal motility, pipeline reduces it through a mechanism that does not appear to involve YR1 (lzzo et at., 2001). The two spicy compounds also appear to affect intestinal harrier function differently: Capsaicin, but not pipeline, increases cell permeability for ions and medium-sized macromolecules (Jensen-Jarolim et at., 1998). Pipeline has also been reported to affect metabolism ofdrugs and other xcnobiotics by decreasing some phase I enzymes (P4502E1). but increasing other phase I enzymes (P4502B and P4501A) as well as phase II enzymes (glucuronidation) (Kang etal., 1994; Shoba et at., 1998).
