One particularly exciting application of the H-2KbtsA58 transgenic mice will be in the generation of cell lines from other strains of mice that are genetically aberrant, either as a result of being bred as a mutant strain of animals or as a result of a mutation introduced by choice (such as mice in which the function of a particular gene of interest has been disrupted, for example). Since cell lines can be generated from heterozygous H-2KbtsA58 transgenic mice, it thus follows that mating of homozygote breeding stock with a homozygous mutant will yield animals expressing both properties of interest. If the mutation of interest is dominantly active, then direct generation of cell lines from F1 litters would be appropriate, whereas recessive mutations would have to be rebred to homozygosity in order for the cell lines of interest to be derived. These strate gies have already been applied to the study of myoblasts from mdx mice, which have a mutation in the dystrophin gene. H-2KbtsA58 x mdx F1 pups have proven to be a ready source of mdx-deficient skeletal myoblast lines, thus providing an important new tool for the study of dystrophin function (78).
